There is a need for comprehensive research on the species structure and the population dynamics of the most common aphidophagous species. A critical factor of the effectiveness of aphid biocontrol is the ratio of beneficial polyphagous (generalist) to oligo- or monophagous (specialist) species within the various trophic groups. Aphids' population density and environmental conditions influence the development and potential feeding of useful insects. The present study aimed to determine the community structure, relationships and diversity between aphids and their aphidophagous species in alfalfa fields using the following methods: sweeping with an entomological net, the quadratic method, coloured sticky board method, route survey method and visual observations. Research on the structure of the aphid–aphidophagous community revealed that aphidophagous species belong to three groups: (1) polyphagous predatory bugs from the families Anthocoridae and Nabidae, (2) oligophagous and polyphagous predators from the families Coccinellidae, Syrphidae and Chrysopidae; and (3) monophagous and oligophagous parasitoids, primarily from the families Aphidiidae and Ichneumonidae. From mid-May to June, there was a sufficiently large potential for aphidophagous species (Coccinellidae, Syrphidae, Chrysopidae, Anthocoridae and Nabidae) to control aphids, while in September, predatory ladybirds from the Coccinellidae family were the main biological control agents. Coccinellidae (Coleoptera) exhibited the highest values of diversity, dominance and richness indices among insect groups in the aphid–aphidophagous community. The existence of diverse aphidophagous species in alfalfa fields suggests that these predators can complement each other, leading to effective biological control of aphids. The synergy among different predator species holds promise for enhancing the overall efficacy of integrated pest management strategies.

As an environmental factor, temperature impacts the distribution of species and influences interspecific competition. The molecular chaperones encoded by small heat shock proteins (sHsps) are essential for rapid, appropriate responses to environmental stress. This study focuses on Hsp20.8, which encodes a temperature-responsive sHsp in Liriomyza trifolii, an insect pest that infests both agricultural and ornamental crops. Hsp20.8 expression was highest at 39℃ in L. trifolii pupae and adults, and expression levels were greater in pupae than in adults. Recombinant Hsp20.8 was expressed in Escherichia coli and conferred a higher survival rate than the empty vector to bacterial cells exposed to heat stress. RNA interference experiments were conducted using L. trifolii adults and prepupae and the knockdown of Hsp20.8 expression increased mortality in L. trifolii during heat stress. The results expand our understanding of sHsp function in Liriomyza spp. and the ongoing adaptation of this pest to climate change. In addition, this study is also important for predicting the distribution of invasive species and proposing new prevention and control strategies based on temperature adaptation.

Triatomines (Hemiptera: Heteroptera: Reduviidae) are hematophagous insects, well-known for their vectorial role in transmitting Trypanosoma cruzi Chagas (Kinetoplastida: Trypanosomatidae) parasites, the etiological agent of Chagas disease. Trapping these insects would limit human-triatomine interaction and, thus, control the disease. In this context, there is a critical need for effective lures to control triatomines. Through double-choice bioassays, we investigated the preference of Triatoma infestans Klug, T. pallidipennis Stal, and Rhodnius prolixus Stal triatomines for: (a) volatiles from fermented products (various fermentation types and substrates) and (b) commercial insect lures. Furthermore, we identified the chemical composition of these volatiles through headspace collection using Solid Phase Micro Extraction coupled with Gas Chromatograph-Mass Spectrometer (HS-SPME-GC-MS). Volatiles from lactic fermentation and certain fermented fruits, along with commercial lures, attracted triatomines, while other products exhibited possible repellent or dislodging properties. These findings hold promise for the control of triatomines and, ultimately, Chagas disease.

The symbiosis between microorganisms and host arthropods can cause biological, physiological, and reproductive changes in the host population. The present study aimed to survey facultative symbionts of the genera Wolbachia, Arsenophonus, Cardinium, Rickettsia, and Nosema in Cotesia flavipes (Cameron) (Hymenoptera: Braconidae) and Diatraea saccharalis (Fabricius) (Lepidoptera: Crambidae) in the laboratory and evaluate the influence of infection on the fitness of these hosts. For this purpose, 16S rDNA primers were used to detect these facultative symbionts in the host species, and the hosts' biological and morphological features were evaluated for changes resulting from the infection caused by these microorganisms. The bacterial symbionts studied herein were not detected in the D. saccharalis samples analysed, but the endosymbiont Wolbachia was detected in C. flavipes and altered the biological and morphological aspects of this parasitoid insect. The results of this study may help to elucidate the role of Wolbachia in maintaining the quality of populations/lineages of C. flavipes.

The Culicidae mosquito family breeds in various natural and artificial water bodies. Phytotelmata are plant structures that hold water and are used by many mosquito species during their life cycle. Mosquitoes are disease vectors, and phytotelmata play an important role in harbouring them. Investigating these phytotelmata will facilitate to develop appropriate site specific vector control strategy. Approximately 750 phytotelmata in 16 locations on Car Nicobar Island was examined. Mosquito larvae were collected from different habitats such as tree holes, root holes, leaf axils, and bamboo stumps. On average, the number of larvae per 10 ml of water ranged from 0.003 to 3.833. Among the 16 mosquito species identified, there were vectors of malaria (Anopheles sundaicus), dengue (Aedes aegypti and Aedes albopictus) and filariasis (Culex quinquefasciatus), belonging to seven genera. The most common phytotelmata were coconut trees (17.5%) followed by Java apple trees (10.3%) and bamboo stumps (7.1%). The most widespread mosquito species was Aedes edwardsi, followed by Aedes malayensis and Ae. albopictus, and the per cent of phytotelmata that contained these species were 42.6, 22.0, and 16.8% respectively. Anopheles mosquitoes had the highest container index (6.1%) in root holes, while Aedes and Culex mosquitoes were more frequently found in bamboo stumps. The alpha biodiversity indices indicated that the mosquito population in Car Nicobar Island was highly diverse (S = 16; DMg = 36.9). Tree holes showed the highest diversity, as all 16 species were collected from them. Assessment of phytotelmata, there is a significant relationship observed between mosquito genus, village, habitat, and tree families, on the remote island of Car Nicobar. Considering the presence of disease-transmitting mosquito vectors in the phytotelmata, these habitats should be considered when implementing control measures to eliminate mosquito-borne infections.

From the 1990s, the Southeast Asia native giant resin bee Megachile sculpturalis (Smith, 1853) was introduced first to North America, and then to many countries in Europe. Despite increasing studies on its invasive potential and geographical expansion, information on nesting behaviour of this species is still extremely scarce. To increase knowledge on the nesting biology of M. sculpturalis, we studied multiple aspects of nesting and pollen provisioning in three consecutive years in artificial nests in Bologna, Italy. We observed 166 bees visiting nests, and followed individual nesting behaviour and success of 41 adult females. We measured cavity diameter in 552 nests and characterised the structure in 100 of them. More than 95% of nest diameters ranged between 0.6 and 1.2 cm, overlapping with several sympatric species of cavity-nesting hymenopterans in the study area. Most nests had a first chamber from the entrance of variable length without brood, followed by an average of about two brood cells with a mean length of 2.85 ± 0.13 cm each. The pollen stored in brood cells was almost monofloral, belonging to the ornamental plant Styphnolobium japonicum (L.) Schott. We estimated that a single female should visit ≈180 flowers to collect enough pollen for a single brood cell. These results fill knowledge gaps on the nesting biology and nest structure of the exotic M. sculpturalis, and they are discussed in relation to possible competition with native bees for nesting sites and foraging resources.

Calling males of Anastrepha obliqua release volatile compounds to attract conspecific males to form leks and females to mate. Male volatiles from Mexican and Brazilian populations of A. obliqua have been previously identified. However, there are differences in the number and identity of volatile compounds between the populations. These differences in volatile profiles may be due to male origin (e.g. wild or mass-reared flies) or methodological issues (e.g. sampling techniques). In this study, we evaluated the attractiveness of wild, laboratory non-irradiated, and laboratory-irradiated flies under semi-field conditions. Male volatiles were collected using dynamic headspace sampling (DHS) and solid-phase microextraction (SPME) techniques, and identified using gas chromatography-coupled mass spectrometry. The results showed no difference in the attractiveness of wild, laboratory non-irradiated, and irradiated males to females. However, the number of captured females differed according to the origin; wild and non-irradiated females were captured more frequently than the irradiated flies. A total of 21 compounds were found using SPME, whereas only 12 were collected using DHS, although the relative amounts of these compounds were higher than those obtained using the former sampling technique. In addition, only laboratory non-irradiated males released α-pinene and menthol, which have not been previously reported in this fruit fly species. Additionally, we identified novel compounds in A. obliqua; however, certain compounds previously reported were not detected. This study suggests that despite the qualitative and quantitative variations in the volatile profiles of A. obliqua males, their attractiveness was unaffected.

The diversity and health of insects that feed on plants are closely related to their mutualistic symbionts and host plants. These symbiotic partners significantly influence various metabolic activities in these insects. However, the symbiotic bacterial community of toxic plant feeders still needs further characterisation. This study aims to unravel bacterial communities associated with the different species of insect representing three insect orders: Thysanoptera, Hemiptera, and Lepidoptera, along with their predicted functional role, which exclusively feeds on latex-rich plant species Ficus microcarpa. By using 16S rRNA gene high-throughput sequencing, the analysis was able to define the major alignment of the bacterial population, primarily comprising Proteobacteria, Firmicutes, Bacteroidota, Actinobacteriota, and Acidobacteriota. Significant differences in symbiotic organisms between three insect groups were discovered by the study: hemipterans had Burkholderia and Buchnera, and lepidopterans had Acinetobacter. At the same time, Pseudomonas was detected in high abundance in both lepidopteran and thysanopteran insects. Furthermore, these symbionts exhibit consistent core functions, potentially explaining how different insects can consume the same host plant. The identified core functions of symbionts open avenues for innovative approaches in utilising these relationships to develop environment-friendly solutions for pest control, with broader implications for agriculture and environmental conservation.

Pebrine disease, caused by Nosema bombycis (Nb) infection in silkworms, is a severe and long-standing disease that threatens sericulture. As parasitic pathogens, a complex relationship exists between microsporidia and their hosts at the mitochondrial level. Previous studies have found that the translocator protein (TSPO) is involved in various biological functions, such as membrane potential regulation, mitochondrial autophagy, immune responses, calcium ion channel regulation, and cell apoptosis. In the present study, we found that TSPO expression in silkworms (BmTSPO) was upregulated following Nb infection, leading to an increase in cytoplasmic calcium, adenosine triphosphate, and reactive oxygen species levels. Knockdown and overexpression of BmTSPO resulted in the promotion and inhibition of Nb proliferation, respectively. We also demonstrated that the overexpression of BmTSPO promotes host cell apoptosis and significantly increases the expression of genes involved in the immune deficiency and Janus kinase-signal transducer and the activator of the transcription pathways. These findings suggest that BmTSPO activates the innate immune signalling pathway in silkworms to regulate Nb proliferation. Targeting TSPO represents a promising approach for the development of new treatments for microsporidian infections.

Phytoplasmas are phloem-limited bacteria that are primarily transmitted by hemipteran insects and are emerging threats to Camptotheca acuminata Decne plants due to their associations with a witches’ broom disease. Despite numerous studies, there has been no report on insect transmission of phytoplasma among C. acuminata. Here, transmission characteristics of the leafhopper, Empoasca paraparvipenis Zhang and Liu, 2008 and the phytoplasma in plant leaves through PCR quantification are described. The interaction between C. acuminata-phytoplasma and insect vectors was examined by analysing the impact on the life characteristics and progeny population in a temperature-dependent manner. Phytoplasma-infected C. acuminata plant exhibited symptoms including shorter internodes, weak and clustered branches, shrunken and yellowed leaves, and red leaf margins. The acquisition and transmission time of bacterial-infected third-instar nymphs of insect vectors were 10 (11.11%) and 30 min (33.33%), respectively. A single insect vector can infect a plant after 72 h of feeding, and the incidence rate of disease increases with the number of insects following 11–100% from single to 20 insects. The development time of the infected insect vectors (1–3 instars) was significantly shorter than that of the healthy insects, and the development duration of instar individuals was longer. In progeny populations, the higher the phytoplasma concentration (88–0% for 1–5 instars nymph, female and male adults), the shorter the development time and the longer the adult lifetime (both male and female). These findings provided research evidence of phytoplasma transmission by insect vectors; however, further investigation of the mechanisms for prevention and management of phytoplasma diseases is needed.

Euwallacea interjectus, a recently discovered pest in poplar plantations, poses a significant economic threat due to its role in causing widespread tree mortality. This pest's cryptic behaviour has hindered research and control efforts, making laboratory rearing a valuable tool for studying its development and biology. We investigated the development period and biological characteristics of E. interjectus using artificial diets and fungal medium. Our findings revealed that the development time for eggs, larvae, and pupae averages approximately 6, 18, and 6 days, respectively. Notably, first and second instar larvae displayed peak moulting periods at 3.45 ± 0.64 SD and 7.92 ± 1.77 SD days, respectively. Furthermore, we measured head capsule widths of postmolt larvae, yielding values of 318.02 ± 7.38 SD μm for first-instar larvae, 403.01 ± 11.08 SD μm for second-instar larvae, and 549.54 ± 20.74 SD μm for third-instar larvae. Our research also uncovered a positive correlation between the number of progeny (eggs, larvae, pupae, and adults) and the mean length of the gallery system. Interestingly, the haplodiploid reproductive strategy did not significantly affect the number of offspring produced by the foundress. Additionally, we observed that foundresses displayed higher fecundity when subjected to nutrient-rich diets as compared to nutrient-poor diets. Our results will deepen our understanding of the biology of E. interjectus and provide criteria for larval instar classification. Additionally, managing nutrient availability within the colony could be considered a viable approach to regulating population size.

The egg parasitoid Anastatus japonicus is a key natural enemy in the biological control of various agricultural and forestry pests. It is particularly used against the brown marmorated stink bug Halyomorpha halys and the emerging defoliator pest Caligula japonica in East Asia. It has been proved that the eggs of Antheraea pernyi can be used as a factitious host for the mass production of A. japonicus. This study systematically documented the parasitic behaviour and developmental morphology exhibited by A. japonicus on the eggs of A. pernyi. The parasitic behaviour of A. japonicus encompassed ten steps including searching, antennation, locating, digging, probing, detecting, oviposition, host-feeding, grooming, and resting. Oviposition, in particular, was observed to occur in three stages, with the parasitoids releasing eggs during the second stage when the body remained relatively static. Among all the steps of parasitic behaviour, probing accounted for the longest time, constituting 33.1% of the whole time. It was followed by digging (19.3%), oviposition (18.5%), antennation (9.6%), detecting (7.4%), and the remaining steps, each occupying less than 5.0% of the total event time. The pre-emergence of adult A. japonicus involves four stages: egg (0 to 2nd day), larva (3rd to 9th day), pre-pupa (10th to 13th day), pupa (14th to 22nd day), and subsequent development into an adult. Typically, it takes 25.60 ± 0.30 days to develop from an egg to an adult at 25℃. This information increases the understanding of the biology of A. japonicus and may provide a reference for optimising reproductive devices.

Bees play a significant role in the health of terrestrial ecosystems. The decline of bee populations due to colony collapse disorder around the world constitutes a severe ecological danger. Maintaining high yield of honey and understanding of bee behaviour necessitate constant attention to the hives. Research initiatives have been taken to establish monitoring programs to study the behaviour of bees in accessing their habitat. Monitoring the sanitation and development of bee brood allows for preventative measures to be taken against mite infections and an overall improvement in the brood's health. This study proposed a precision beekeeping method that aims to reduce bee colony mortality and improve conventional apiculture through the use of technological tools to gather, analyse, and understand bee colony characteristics. This research presents the application of advanced digital image processing with computer vision techniques for the visual identification and analysis of bee brood at various developing stages. The beehive images are first preprocessed to enhance the important features of object. Further, object is segmented and classified using computer vision techniques. The research is carried out with the images containing variety of immature brood stages. The suggested method and existing methods are tested and compared to evaluate efficiency of proposed methodology.

Since metabolism, survival, and reproduction in hexapods are closely related to temperatures; changes in the mean and variance of temperature are major aspects of global climate change. In the typical context of biological control, understanding how predator–prey systems are impacted under thermal conditions can make pest control more effective and resilient. With this view, this study investigated temperature-mediated development and predation parameters of the predator Harmonia axyridis against the potential prey Spodoptera litura. The age-stage, two-sex life table of the predator was constructed at four temperatures (i.e. 15, 20, 25, and 30°C) by feeding on the first instar larvae of S. litura. Our results showed that the mean generation time (T) decreased but the intrinsic rate of increase (r) and the finite rate of increase (λ) increased with increased temperature. The mean duration of the total preadult stage decreased with higher temperatures. The T and r were 70.47 d and 0.0769 d−1 at 15°C; 58.41 d and 0.0958 d−1 at 20°C; 38.71 d and 0.1526 d−1 at 25°C; and 29.59 d and 0.1822 d−1 at 30°C, respectively. The highest net reproductive rate (R0) and fecundity were obtained at 25°C. The highest λ (1.1998 d−1) and lowest T (29.59 d) were obtained at 30°C, whereas the maximum net predation rate (C0) was at 25°C. Total population and predation rates projections were the highest at 30°C. Based on these findings, we anticipate that biological control strategies for this predator release against S. litura should be attuned to warming scenarios to achieve better biocontrol functions.

Peach-potato aphids, Myzus persicae Sulzer (Hemiptera:Aphididae), and cabbage aphids, Brevicoryne brassicae Linnaeus (Hemiptera:Aphididae), are herbivorous insects of significant agricultural importance. Aphids can harbour a range of non-essential (facultative) endosymbiotic bacteria that confer multiple costs and benefits to the host aphid. A key endosymbiont-derived phenotype is protection against parasitoid wasps, and this protective phenotype has been associated with several defensive enodsymbionts. In recent years greater emphasis has been placed on developing alternative pest management strategies, including the increased use of natural enemies such as parasitoids wasps. For the success of aphid control strategies to be estimated the presence of defensive endosymbionts that can potentially disrupt the success of biocontrol agents needs to be determined in natural aphid populations. Here, we sampled aphids and mummies (parasitised aphids) from an important rapeseed production region in Germany and used multiplex PCR assays to characterise the endosymbiont communities. We found that aphids rarely harboured facultative endosymbionts, with 3.6% of M. persicae and 0% of B. brassicae populations forming facultative endosymbiont associations. This is comparable with endosymbiont prevalence described for M. persicae populations surveyed in Australia, Europe, Chile, and USA where endosymbiont infection frequencies range form 0–2%, but is in contrast with observations from China where M. persicae populations have more abundant and diverse endosymbiotic communities (endosymbionts present in over 50% of aphid populations).

The European grapevine moth Lobesia botrana (Denis & Shiffermüller 1776) is an economically important pest of the vine-growing areas worldwide. Chemical insecticides have been used for its control; however, its resistance status is largely unknown in many regions. We monitored the susceptibility of several L. botrana populations from Greece and Turkey. In addition, based on RNAseq transcriptome analysis, we identified and phylogenetically classify the cytochrome P450 genes of L. botrana, as well as analysed target site sequences and looked for the presence of known resistance mutations. Resistance against chlorantraniliprole, alpha-cypermethrin, spinetoram, etofenprox, and acetamiprid was very low (below 2.5-fold in all cases, compared to a reference strain from Greece) in all populations from Greece that were included in the study. However, resistance against indoxacarb (4–30-fold), spinosad (5–59-fold), and deltamethrin (18–30 fold) was detected in the L. botrana populations from Turkey, compared to a reference population from Turkey. De novo transcriptome assembly and manual annotation, and subsequent PCR-based analysis of insecticide target sequences (i.e. voltage-gated sodium channel – VGSC: target of pyrethroids and oxadiazines; nicotinic acetylcholine receptor subunit a6 – nAChR_α6: target of spinosad; ryanodine receptor – RyR: target of diamides; glutamate-gated chloride channel – GluCl: target of avermectins and; acetylcholinesterase – AChE: target of organophosphates) showed the absence of known resistance mutations in all specimens from both countries. Finally, the L. botrana CYPome (116 genes) was manually analysed and phylogenetically characterised, to provide resources for future studies that will aim the analysis of metabolic resistance.

Dietary restriction-influenced biological performance is found in many animal species. Pardosa pseudoannulata is a dominant spider species in agricultural fields and is important for controlling pests. In this study, three groups – a control group (CK group), a re-feeding group (RF group), and a dietary restriction group (RT group) – were used to explore development, mating, reproduction, and the expression levels of Vg (vitellogenin) and VgR (vitellogenin receptor) genes in the spider. The findings indicated that when subjected to dietary restriction, the carapace size, weight of the spiderlings, and weight of the adults exhibited a decrease. Furthermore, the preoviposition period and egg stage were observed to be prolonged, while the number of spiderlings decreased. It was also observed that re-feeding reduced cannibalism rates and extended the preoviposition period. Dietary restriction also affected the expression of the Vg-3 gene in the spider. These results will contribute to the understanding of the impact of dietary restriction in predators of pest control, as well as provide a theoretical foundation for the artificial rearing and utilisation of the dominant spider in the field.

Confronting environments with low relative humidity is one of the main challenges faced by insects with expanding distribution ranges. Anastrepha ludens (the Mexican fruit fly) has evolved to cope with the variable conditions encountered during its lifetime, which allows it to colonise a wide range of environments. However, our understanding of the mechanisms underpinning the ability of this species to confront environments with low relative humidity is incomplete. In this sense, omic approaches such as transcriptomics can be helpful for advancing our knowledge on how this species copes with desiccation stress. Considering this, in this study, we performed transcriptomic analyses to compare the molecular responses of laboratory-reared A. ludens exposed and unexposed to desiccation. Data from the transcriptome analyses indicated that the responses to desiccation are shared by both sexes. We identified the up-regulation of transcripts encoding proteins involved in lipid metabolism and membrane remodelling, as well as proteases and cuticular proteins. Our results provide a framework for understanding the response to desiccation stress in one of the most invasive fruit fly species in the world.

Negative relationships between the parental age and offspring life history traits have been widely observed across diverse animal taxa. However, there is a lack of studies examining the influence of parental age on offspring performance using mites, particularly phytoseiid predators as subjects. This study explored the influence of maternal age on offspring life history traits in Amblyseius herbicolus (Chant) (Acari: Mesostigmata), a phytoseiid predatory mite reproducing through thelytokous parthenogenesis. We hypothesised that increased maternal age negatively impacts offspring traits, including developmental duration, body size, fecundity and lifespan. Amblyseius herbicolus was reared under controlled laboratory conditions, and the life history traits of offspring from mothers of varying ages were analysed using linear mixed-effect models. Our results showed that the increase in maternal age significantly reduced individual egg volume, but did not significantly affect offspring developmental duration, body size, fecundity or lifespan. These findings indicate that while older A. herbicolus females produced smaller eggs, the subsequent performance (i.e. body size, fecundity and lifespan) of offspring remained largely unaffected, suggesting possible compensatory mechanisms in the offspring or alternative maternal provisioning strategies. The results of this study offer useful insights into the reproductive strategies of phytoseiid predators and asexually reproducing species, enhancing our understanding of how maternal age affects offspring fitness. Further studies can examine how offspring of A. herbicolus from mothers of different ages perform under adverse environmental conditions.

Peatlands, shaped by centuries of human activities, now face a primary threat from mining activities. Vulnerable to drainage and hydrological instability, peatland areas encounter challenges that compromise their ecological integrity. This study hypothesised that permanent water reservoirs within mines could serve as refugia for water beetles from adjacent areas prone to drying in the summer. Employing standard methods, including entomological scraping and water traps, samples were collected. Results revealed that, in most cases, water beetles exhibited a preference for the Nature 2000 area untouched by mining. Despite unfavourable conditions, the Nature 2000 area showcased a more diverse water beetle fauna. Remarkably, the selected Nature 2000 area, despite its identified degradation based on flora, remained a biodiversity hotspot for peatland water beetle fauna. The study underscores the significance of assessing insects, particularly beetles, as rapid responders to environmental changes. This evaluation holds crucial implications for peatland restoration planning and decision-making regarding mining investments in proximity to peatland areas.
