Maize (Zea mays L.) is one of the main staples in the Peruvian Andes. Phenotypic diversity is extremely high in terms of plant morphology, grain colour, grain size and uses. However, little is known about the genetic component of the Peruvian maize. Genotyping-by-sequencing was used, for the first time, to infer the genetic diversity and population structure of 25 accessions of maize collected from two ecoregions of the departments of the Peruvian highlands, Cajamarca and Huancavelica, to provide a basis for the conservation and breeding of this crop genetic resource. A total of 29,332 single-nucleotide polymorphisms were identified along all 10 chromosomes of maize. STRUCTURE analysis revealed two clusters (open-pollinated cultivars and landraces). Principal coordinate analysis and our dendrogram showed that these accessions of Peruvian maize do not group based on their geographic origin but on their improvement status, however, this is weakly supported. Average genetic diversity was very high (0.35). Analysis of molecular variance showed a reduced variation between populations (2.85%) and indicated that variability within populations is 97.15%. The lowest population divergence was zero for populations of Colcabamba versus Pichos and Daniel Hernandez versus Colcabamba and the highest population divergence (0.041) was observed for population Cajamarca versus Santiago de Tucuma. As expected, FIS values were positive. Additional samples from other Andean and west coast Peruvian localities are needed to provide a better understanding of the genetic components of this important crop aiming to develop a modern breeding programme of maize in Peru.

The genetic diversity of Cyclanthera pedata was assessed through agro-morphological, mineral composition and biochemical traits. Field surveys across India documented its wide ecological distribution (18.66°–31.02°N, 78.05°–83.27°E, 131–3665 m AMSL) in Andhra Pradesh and northwest Himalayan region. Field evaluations were conducted during Kharif 2021 and 2022 at ICAR-Vivekananda Parvatiya Krishi Anusandhan Sansthan, Almora (1250 m AMSL). Significant genotypic variation was observed in fruit yield (195–1505 g/plant), fruit numbers (36–364 per plant), chlorophyll content (0.49–2.61 μg/g) and antioxidant activities (16.02–39.04 mM Trolox equivalent/g DW). Notable genotypes included RK-40 (highest yield of 1505 g/plant), RK-01 (early flowering) and RK-33 (early fruiting). High coefficients of variation (>23%) were noted in total chlorophyll, carotenoids and FRAP values. Genotype RK-42 excelled in antioxidant activities and carotenoids. Hierarchical cluster analysis grouped the germplasm into four clusters based on critical traits independent of geographic origin. In contrast, principal component analysis identified traits like polyphenols, antioxidants, seed weight and fruit yield as significant contributors to phenotypic variation. The biplot revealed relationships between traits, such as positive correlations between fruit yield, fruit and pulp weight, and biochemical traits like DPPH and ABTS. Genotypes such as IC 430006, IC 356007, IC 393261, IC 393308, UKAl 1 and IC 361297 display distinct profiles. These findings highlight critical traits for breeding and conservation efforts, particularly for enhancing fruit yield and biochemical attributes in C. pedata germplasm.

The present study was conducted with the objective of studying the genetic diversity of essential oils (EOs) in curry leaf (CL) ex-situ. Chemometric methods and pattern analysis were employed to assess the genetic diversity of EOs and to characterise diverse sets of CL germplasm into different chemotypes. The study revealed a huge genetic diversity for EO yield and its composition among the tested genotypes. Cultivated types had significantly higher EO yields and showed a greater degree of genetic divergence compared to wild types. In total, 80 different compounds were identified in the EOs of CL and classified into major (6) and minor (74) compounds. The major compounds α-pinene, γ-terpinene, and α-selinene and 14 minor compounds were highly variable among the tested genotypes. They may play an important role in the formation of different chemotypes. Other important compounds, such as trans-caryophyllene and α-humulene, were more widely distributed among the tested genotypes and indicated their predominant occurrence in the EOs of CL. Some major compounds, such as valencene and γ-terpinene, showed a significant regional correlation, indicating the role of geographic factors in the evolution of different chemotypes. Furthermore, some compounds such as α-pinene, bornyl acetate, and camphene had significantly higher concentrations in wild types compared to cultivated types, indicating the influence of domestication through human selection on the composition of EOs in CL. A total of 4 major chemotypes were characterised, of which three new chemotypes are being reported for the first time in CL.

A set of 68 simple sequence repeat (SSR) markers were selected from existing databases (including Medicago, soybean, cowpea and peanut) for the purpose of exploiting the transferability of SSRs across species and/or genera within the legume family. Primers were tested for cross-species and cross-genus fragment amplification with an array of 24 different legume accessions. Nearly one-third (30.78%) of the SSR primers screened generated reproducible and cross-genus amplicons. One hundred and seventeen cross-species polymorphic amplicons were identified and could be used as DNA markers. These polymorphic markers are now being used for characterization and evaluation of our collected and donated legume germ- plasm. The transferability of SSRs, mis-/multiple-primings, homologous/heterologous amplifications, single/multiple-amplicons and application of these amplicons as DNA markers are discussed. The transfer of SSR markers across species or across genera can be a very efficient approach for DNA marker development, especially for minor crops.

Peanut (Arachis hypogaea L.) is an important row crop rich in oil, protein, vitamins and other micro-nutrients. The intensive selection of the cultigen, a cultivated plant deliberately altered by humans through cultivation, has resulted in favourable changes in yield and biochemical composition. Nevertheless, it has generated a narrow genetic basis that limits the development of new varieties with resistance to pests, diseases and environmental stresses. In this study, we address this limitation by characterizing the proximate and fatty acid composition of a multi-disease-resistant interspecific recombinant inbred line (RIL) population derived from three wild Arachis species and a cultivated elite peanut line that is being used to widen the genetic basis of the crop. The population was also genotyped with the Axiom Arachis 48K SNP array and used to detect quantitative trait loci (QTL) for oil, protein content and oleic and linoleic fatty acid percentages. A wide range of proximate composition was found in the RIL population. Eighteen and 11 individuals had high oil and protein content, respectively, and no undesirable traits related to oil quality had been introduced into the population from wild species. The fatty acid composition of oleic and linoleic acids was found to be regulated by two major QTL. The discovery of markers within the major effect QTL for the most significant chemical traits provides new opportunities for the creation of resistant and extremely nutrient-dense peanut cultivars.

Preference for functional and nutritious food capable of meeting consumers' demand and health is on the increase. The present preliminary study seeks to assess physico-chemical and nutraceutical diversity in the cocoa bean powder of 77 genotypes present in four Nigerian cocoa field banks. Twenty ripe pods/genotypes in each of the four active breeding field banks at the Cocoa Research Institute of Nigeria (CRIN), Ibadan, Nigeria were utilized. Composite beans from the 20 pods of each genotype were singly fermented, sun-dried and milled. Duplicate samples of the powder of each genotype were analysed for physico-chemical and nutraceutical components. Twenty-one polymorphic variables distinguished the 77 cocoa genotypes. Grouping by dendogram identified four clusters, three differently and uniquely captured 100% of the genotype membership in the local clone, international clone and the regional varieties field bank but 86% of the genotypes in the hybrid trial field bank were grouped in cluster I. Prominent traits with highest values in each clusters were: protein, pH, Ca, K and Fe (Cluster I), Zn and Mg (Cluster II), crude fat and P (Cluster III) and crude fibre, ash, theobromine, flavonoids and caffeine (Cluster IV). Exploitable diversity for nutritional quality improvement is present in the active breeding and working collections of Nigerian cocoa field banks.

Twenty-nine exotic common bean germplasms and three elite cultivars were examined for phenotypic diversity in two bean-producing environments (Kanpur and Shimla) across three winter seasons and one rainy season. The estimate of genetic variability parameters revealed that the exotic bean germplasm has enough diversity for all the evaluated features. The highest genotypic and phenotypic coefficients of variation were found in seed yield, followed by 100-seed weight, pods per plant and pod length. Furthermore, seed yield was the most heritable and genetically advanced quantitative feature, followed by 100-seed weight, pod length and pods per plant. According to a trait association study, the days to maturity of phenological traits have a strong positive correlation with the days to initial flowering and the days to 50% flowering. Pods per plant and seeds per pod most strongly influence increased grain yield. The first two principal components accounted for 63.3% of the variation and demonstrated significant diversity among exotic bean lines for the traits studied, according to the principal component analysis. According to the hierarchical clustering analysis, 29 accessions and three cultivars were divided into three groups. Cluster I contains early flowering and maturing accessions, while cluster III contains high pods per plant and an increased grain yield of germplasms. The fundamental source of phenological fluctuations in both environmental circumstances is temperature. This study found four genetically divergent and stable performance accessions, including EC932021, EC932189 (earliness), and EC931452, EC931971 (high grain yield), which may aid in the establishment of a bean breeding programme.

Ever increasing global population necessitates the need to increase availability of affordable foods with high nutritional quality. Chickpea being the second most important grain legume cultivated worldwide has the great potential to alleviate problems of protein malnutrition and hidden hunger. The objective of the present investigation was to characterize a diverse set of 402 germplasm comprising of more than 100 commercial cultivars widely cultivated and utilized by breeders for grain protein in two contrasting environments to explore the magnitude of natural genotypic diversity present in the cultivated chickpea and the scope for enhancing it and its quality through breeding. The cultivars containing more than 25 to 30.44% grain protein have been found in the study and large scale production of such protein rich cultivars is expected to increase availability as well as consumption of high quality chickpea necessary to overcome all forms of malnutrition. Trait specific germplasm containing grain protein up to 33.56% are also identified for their further utilization as potential donors in the chickpea improvement programme. The amino acid profiling of selected high and low protein containing genotypes showed considerably greater average amino acid score for all the essential amino acids than the WHO recommended requirement values and is comparable to those of the ‘complete protein’ sources of food suggesting that chickpea deserves to be considered as a ‘complete protein’ source.

Horseradish is a crop grown for its edible underground parts. The development of new cultivars is hindered by the species' predominant vegetative reproduction, making it essential to evaluate locally cultivated accessions to identify new types suitable for cultivation. To this end, 11 horseradish accessions from family vegetable gardens in Trentino-Alto Adige, Italy were examined using 26 qualitative and six quantitative morphological descriptors and characterized by the five major glucosinolates (GSLs) present in the rhizome compared to a reference cultivar. A wide range of variability was observed for the considered qualitative morphological traits. The rhizome's top and basal diameters were 9.9 and 6.2 cm, respectively, with an average fresh weight of 521 g. Total GSL content ranged between 79.5 and 133.5 μmol/g dry weight (DW), with sinigrin (SIN) being the primary component at an average content of 110.0 μmol/g DW. Differences among the investigated accessions were noted for quantitative traits describing their productive features and for GSL content. A positive correlation was discovered between the biometric traits of the plant's underground parts and the SIN and total GSL content, suggesting a link between the quality and yield of the edible product. According to the multivariate analysis, accessions were grouped into three main clusters: the largest of the reference cultivar and the majority of accessions with similar productive and qualitative traits; another featuring two with good qualitative and productive characteristics. The investigated accessions proved to be a valuable germplasm source for cultivating the species.

Chenopodium quinoa W. is a species of South America with an exceptional nutritional content and wide agroclimatological adaptation. It has great genetic and phenotypic variability, however in Colombia there are few genetic improvement programmes that take advantage of its great genetic and productive potential. In Cundinamarca there are some adapted genotypes which have been selected by farmers. We evaluated 36 genotypes of Blanca de Jericó, Blanca Subachoque, Aurora, Púrpura and Tunkahuan from Cundinamarca, using eight ISSR markers. The analysis by the coefficient of Nei-Li at the level of similarity of 0.40 divided the population into three groups according their background genetic and the colour of oxalates. The percentage of polymorphic loci was higher than 90%. The average value of heterozygosity was 0.32, which is low given the selection processes that the evaluated germplasm has undergone. We found moderate genetic differentiation (Fst = 0.23). The analysis of molecular variance (AMOVA) showed higher variation (77%) groups than among groups (23%). The results revealed intra-population diversity, which suggests that farmers within their farms should undergo a more rigorous seed selection process. Our results demonstrate that ISSR markers are useful and powerful to assess the genetic relationships, polymorphism and genetic diversity of quinoa cultivars. The genetic characterization results reported in the present study will be promising for guiding the breeding of quinoa seed quality in Colombia.

Assessing genetic diversity and identifying trait-specific germplasm within germplasm collections is necessary for a varietal development programme. Agronomic features were investigated in 318 diverse dry bean germplasm accessions, including check varieties. We observed a lot of genetic variability for the traits studied. A wide range of variations was noticed for days to 50% flowering, days to maturity, pod length, the number of seeds per pod and 100-seed weight (HSW). For eight of the agronomic features evaluated, the analysis of variance revealed substantial differences among the accessions. For all characters, phenotypic coefficient of variation estimations were more significant than genotypic coefficient of variation. Plant height, days to 50% flowering, seed yield (q/ha) and HSW had high heritability and genetic advance as a per cent of the mean. Association analysis revealed a significant positive relationship between HSW, plant height, pod length and seed yield (q/ha). According to a hierarchical clustering analysis based on agronomic features, the diversity of dry bean germplasm has no significant association with their geographical origin. The number of pods per plant, plant height, days to maturity, days to 50% flowering and seed yield had relatively long vectors based on principal components 1 and 2, indicating that genotypes differ significantly. Additionally, the trait-specific donors and bean common mosaic virus disease-resistant accessions, IC360831, ET4515, EC150250, IC340947, IC564797B, EC565693 and ET8409 could be of value for dry bean improvement.

A study was conducted to collect and characterize the indigenous Kala zeera (Bunium persicum Boiss. Fedtsch.) accessions from the hills of northwestern Himalayan sates of Jammu and Kashmir, Himachal Pradesh and Uttarakhand. Around 1000 accessions were collected during this exploration mission, out of which diversity of 252 accessions was established through morphological characterization. The morpho-agronomic characterization and the analysis of trait data revealed significant variability in number of branches per plant, number of umbelets per umbel, number of seeds per plant, seed yield per plant and 1000 seed weight. The collection and characterization of these diverse accessions can prove useful in future Kala zeera improvement programmes in the world as this is the first such comprehensive report of the crop from northwestern Himalayan region of India.

In Morocco, sugar-beet seed production represents a new challenge to meet the main breeding programme goals. The identification of a suitable zone for plant vernalization represents a bottleneck for seed production. This study aimed chiefly at evaluating the vernalization aptitude of 18 sugar beet half-sib progenies. Trials were conducted during three selection cycles in a specifically chosen environment. The experimental site of Merchouch is 40 km south-east of Rabat city. Field trials design is according to an open-pollinated experimental block. Yield components and five phenological traits were recorded during the growing cycle and at the plant maturation. The variance analysis showed a significant effect of the selection cycle on the phenological traits and grain yield. The plant cycle duration increased by 10.7 days between the first and the third selection cycles. According to the principal component analysis and the hierarchical clustering, F2, F5, F6, F9, F10, F11, F14 and F15 half-sib progenies are described as long cycle and high grain yield families. Grain yield reached 257.3 g per plant, the duration to maturity 350.6 days and the germination rate 93%. The three selection cycles and the suitability of the site vernalization conditions are potentially effective for seed production progress. Our results are relevant as they established an adequate site for the sugar beet seed production. The half-sib selection is a valuable method for sugar beet germplasm enhancement.

The cryopreservation of murine ovarian tissue and its transplantation can be a promising technique for the preservation of fertility and an alternative for the future reconstitution of scientific valuable strains of mice. Accordingly, the aim of this study was to describe the entire surgical procedure for ovariectomy and dorsal subcutaneous autotransplantation in mice, and also some data about the efficiency of this procedure. Female C57Bl/6J mice (n = 18) were anaesthetised and bilaterally ovariectomized. After surgery, ovaries were autotransplanted in small subcutaneous pouches in the dorsal region of the forelimbs. The animals were inspected daily and, 23 days after transplantation, euthanasia and recovery of ovarian tissues were performed. Postoperative recovery, oestrous cyclicity, and folliculogenesis progression were evaluated. At 23 days after transplantation, the recovery of the ovaries was feasible, all classes (primordial to antral) of follicles were observed. Additionally, satisfactory efficiency rates were obtained, with 100% of anaesthesia survival rate, survival, graft recovery, folliculogenesis progression and oestrous cyclicity. In general, this short article describes ovarian ectopic autologous transplantation as an effective technique for maintaining rodent oogenesis and endocrine ovarian function. Even more broadly, we can still assume that the application of this technique may reduce the number of breeding matrices and experimental animals in the near future.

This study assessed the genetic diversity of African yam bean (AYB) accessions using morphological and molecular markers. The accessions were grown, and morphological data collected were subjected to analysis of variance and multivariate analyses. Genomic DNA extracted from the accessions were amplified with inter simple sequence repeat (ISSR) markers. The diversity analysis was conducted using MEGA4 software. The accessions varied significantly (P < 0.05) in growth, flowering and seed-related parameters. Flowering commenced early in most accessions. Weight of 100-seed range from 15.01 to 21.15 g with the mean value of 18.30 g. Significant correlations existed between stem height, the number of leaves and leaf dimensions. Also, days to flowering correlated with pod formation; likewise, seed dimension had a positive association with seed weight. The principal biplot revealed that two components accounted for 41.77% of the observed variation. Analysis of the electropherogram showed 95 loci comprising 1351 alleles were detected by the ISSR markers with 65.26% polymorphism and combined polymorphic information content of 0.85. The principal coordinate analysis placed accessions together on a plane based on their spatial relationship. The dendrogram showed accession pairs (TSs-77, TSs-95) and (TSs-111, TSs-84) are closely related. The phylogram identified three kinships with a total length of 454. Accession TSs-115 is likely the progenitor while TSs-82 and TSs-86 are the most recent. The study concluded that a combination of morphological and ISSR markers is effective for the diversity study of AYB and the existing, genetic diversity in the accessions could be harnessed for its improvement, conservation and utilization.

Guinea grass (Megathyrsus maximus Jacq.) is an important forage species in vast rangelands/grasslands of India and several tropical countries owing to its high biomass yield, good nutritional quality and wide adaptation. Evaluation of the existing natural variation and selection of desirable genotypes is the most plausible breeding method for this apomictic and polyploid grass. Developing a core sub-set to narrow down the number of germplasm required for future genetic studies is also pertinent. The present study involved characterization of 152 diverse M. maximus germplasm representing collections from different agro-ecological zones of India as well as those procured from Africa and Brazil; and development of a core sub-set. Nineteen metric, seven non-metric and nine nutritive traits together established the presence of wide variability among the genotypes. Clustering of the genotypes resulted in eight distinct clusters. The largest cluster included genotypes from Ethiopia, north India, north-western India, south India and north-eastern hill region, thus represented the highest diversity. Eleven of the total 26 Ethiopian genotypes clustered together. Non-metric morphological traits effectively differentiated the genotypes, and were associated with nutritional quality also. Genotypes which flowered once in a year showed slightly better crude protein and digestibility. The clusters were further sub-clustered and representatives were selected to develop the core sub-set of 23 genotypes comprising 20 indigenous and three exotic accessions. Comparison of the range of diversity and mean value for traits as obtained in the core sub-set and that in the total germplasm indicated successful capturing of maximum diversity in the core sub-set.

Cordia sinensis, locally known as ‘Goondi’ in India, is an underexploited multipurpose fruit species found in hot arid regions that is well adapted to drought, salt and hot conditions. The present study was undertaken to collect fruit samples from different locations in the Kachchh region of Gujarat, India, and to determine their field establishment for characterization, conservation and utilization. The maximum distribution of the species was observed in Bhuj (45%) and Mandvi (25%). Field boundaries (35%) and scrub forests (30%) had greater frequencies, whereas backyards had rarer frequencies (10%). The species most commonly occurred on levelled topography (60%) with a soil pH in the range of 8–8.5 (63%). Morphological data of three-year-old plants in the field gene bank showed a maximum coefficient of variation in the number of leaves per plant (66.6), followed by the number of branches per plant (45.62) and collar diameter (27.69). Wide variations were recorded in plant height (121.67–212 cm), spread (118–223 cm2) and the number of branches per plant (6–24.33). Specific accessions were identified for fodder (CBCG-12, CBCG-13 and CBCG-16), early flowering and fruiting (CBCG-12, CBCG-13 and CBCG-14), easier propagation by seeds (CBCG-12 and CBCG-13) and salt tolerance (CBCG-15 and CBCG-16). Preliminary findings and information provided about this species' utilization and other aspects might be useful for future research on its domestication, sole plantation and conservation aspects, improving the exploitation of this species by present and future generations.

Bread wheat (Triticum aestivum) is one of the most important food crops in the world. Its physiological and morphological traits are closely related to yield. Therefore, it is generally important to discover the genomic region associated with these traits. In this research, associations between 21 simple sequence repeat (SSR) markers and 10 inter-simple sequence repeat (ISSR) markers with some traits related to root structure at the embryonic and seedling stages and also some agromorphological traits at the whole plant stage were evaluated on a set of 102 wheat genotypes. A highly significant coefficient of variation among different genotypes was observed in all measured traits. A high level of polymorphism with SSR and ISSR markers was obtained. Genetic structure analysis revealed two distinct subpopulations. Significant correlations were found between genomic markers and evaluated traits. A total of nine markers, including four SSR markers in 1A, 3A, 5A and 2B chromosomal regions and five different ISSR markers were related to the studied traits. Several molecular markers were significantly associated with more than one phenotypic trait, suggesting the possible presence of pleiotropic or indirect effects. The phenotypic variation justified by these alleles ranged from 4 to 15%. Obtained genetic information can be targeted for further validation and genetic analysis in the relevant populations or other breeding sets.

Cotton is one of the most important crops among natural fibres. Fibre quality determines the spinning ability, which is negatively correlated with yield and yield-contributing traits. Limited efforts have been made to improve fibre quality and yield in diploid cotton. Therefore, screening a large panel of germplasm lines can help identify genotypes with better fibre quality and yield. We evaluated 712 desi cotton genotypes for fibre quality traits. The genotypes showed a significant difference for all the traits, suggesting considerable variability for fibre quality improvement. Fibre length and strength showed high phenotypic and genotypic coefficients of variation. Heritability was high for fibre strength, length, and elongation. Fibre length and strength were positively correlated; however, micronaire was negatively correlated with these two traits. Superior accessions were identified for fibre length (11), strength (20), uniformity (7), and elongation (25) among genotypes. Most of the desi lines (71%) had medium micronaire values. Twenty accessions identified were ideal for spinning, showing the fibre strength-to-length ratio of one. Cluster analysis based on Euclidean distance grouped all 712 accessions into four major clusters. Principal component analysis biplot revealed that accessions AC3418, 360-SP1, AC3522B, Kanpur A, Gao16CB-9, and AC3370 were genetically diverse. The superior accessions for fibre quality identified in this study are potential lines for the diploid cotton improvement programme.

The current study was undertaken to identify the sources of tolerance to bruchid in cowpea, by screening a set of germplasm accessions as a source for natural resistance. A total of 103 diverse accessions of cowpea were evaluated for resistance to Callosobruchus maculatus Fab. under no-choice artificial infestation conditions. Significant differences among the cowpea accessions were observed for oviposition, adult emergence, exit holes and per cent seed weight loss (PSWL) caused by the bruchid infestation. The accessions showed variation in physical seed parameters viz., colour, shape, testa texture, length, width and seed hardness. Among the seed biochemical parameters studied, per cent sugar content ranged from 0.322 (IC330950) to 1.493 (IC249137), and per cent phenol content ranged from 0.0326 (EC390261) to 1.081 (EC528423). Correlation studies indicated that PSWL had significant positive correlation (r = 0.335) with exit holes, oviposition (r = 0.219), adult emergence (r = 0.534) and seed roundness (r = 0.219). Adult emergence had a significant negative correlation with seed hardness (r = −0.332). Correlation with biochemical parameters indicated that PSWL had a significant positive correlation (r = 0.231) with sugar content and a significant negative correlation with phenol content (r = −0.219). None of the accessions were found to be immune to bruchid infestation. However, out of studied accessions, EC528425 and EC528387 were identified as resistant based on PSWL and moderately resistant based on adult emergence. These resistance sources of cowpea germplasm can be used as potential donors for development of bruchid tolerant/resistant cultivars.
